The caulking on my roof around the plumbing vent and fan vent is cracking.Any thouts

scrape it off and use some good outdoor caulk. this stuff is good, from lowes.

No.. Around those areas you use self leveling sealant, or "lap sealant". Just wipe down what is there with some alcohol to clean it up, and lay new over. Unlike silicone, lap sealant will stick to itself.

Is it best to remove the old caulking first and apply the new, or is it sufficient to apply over the old stuff. If that's the case should there be any prep of the old caulking first?

When dealing with the areas that have caulk, you must remove the old. All of it. It won't stick to itself. But lap sealant is a whole other critter. It will stick to itself, and remains pliable and sticky for years.. heck, decades.
Just a couple weeks ago I picked a little wad out of my shoe tread, that was pulled from the roof on our '79 Airstream, about 6 years ago, and has been lurking in the gravel driveway just waiting for a chance to attack. As far as I could tell, it had been applied to the camper when new.
