My wife and I are heading up to Glendo State Park next week for some mountain biking. Then we'll stay two nights at Keyhole State Park near Devil's Tower and back home by way of Chadron State Park in Nebraska for some more biking. Looking forward to a short vacation and some new sights to see! Any recommended must do activities for this region?

I highly recommend Fort Robinson state park, west of Chadron. 
You ought to love Glendo, most of the campers/boaters there are from Colorado Smile   Avoid it on the weekend, if possible.

I there is a fed park at Devils tower. also wind cave fed park in S. Dakota

We just returned from a short vacation to the Black Hills - towed our 178 from the St Louis area. I'm retired AF so we stayed at the 'famcamp' at Ellsworth AFB (Rapid City, SD) and from there made trips to the Bad Lands (1hr), Crazy Horse (< 1hr), Mt Rushmore (.5 hr), Custer State Park (.5hr), Devils Tower (2hr). Keystone and Deadwood were packed with tourists so we drove through, didn't stop. We traveled through SD on I90 and stopped at Wall Drug (Wall, SD), 1880 Town (Midland SD), and St Joseph college - Indian museum and cultural museum (Chamberlin, SD). Going to the Black Hills we spent a night in Sioux Falls at the KOA (well run), and on the return Omaha, NE KOA (adequate, but really packed). We particularly liked Crazy Horse (participated in the 10k Volksmarch), Bad Lands, and Devils Tower - probably make another trip out in that direction in a few years and include Yellow Stone. How did you find this camp? I am also retired mil and looking for a list of "famcamps" around the nation.
